Cytotoxic Effects of Ferula Latisecta on Human Glioma U87 Cells

Abstract: Glioblastoma multiforme (GBM) is the fatal type of astrocytic tumors with a survival rate of 12 months. The present study, for the first time, evaluated the cytotoxic impacts of Ferula latisecta (F. latisecta) hydroalcoholic extract on U87 GBM cell line. The MTT assay measured the cellular toxicity following 24- and 48 h treatment with various doses of F. latisecta (0–800 μg/mL). Apoptosis was evaluated by an Annexin V/propidium iodide (PI) staining 24 h after treatment by F. latisecta. Moreover, to determine … Show more

“…It was evidenced that LnHS at dose levels higher than 25 µg/mL, induced clear cell morphological changes as cells shrunk and loss cell adhesion ( Figure S2). Thus, in order to better understand and define a preliminary LnHS-cell death inducing mechanism, the effects on cytotoxicity was further assessed on U-87 glioblastoma cells, which are characterized by a rapid migration, and a highest invasion capacity [48]. Human Fibroblast (HF) cells were used in order to evaluate LnHS effects on non-tumorigenic cells.…”

“…As previously described, a colorimetric MTT assay was used to assess cell proliferation [4,5]. Briefly, U-87 cells were seeded in 96 wells (10 4 per each well), and after overnight, the cells were incubated with various concentrations of Zerumbone (0-400 μM) for 24 and 48 hours.…”

“…GBM is characterized by increased proliferation and a lack of apoptosis [3]. Given extensive treatments, including maximal resection, radiation, or che-motherapy with the alkylating agent temozolomide (TMZ), patients of GBM typically have an abysmal prognosis [4]. Hence, this emphasizes the necessity of new treatment approaches in GBM patients.…”

“…As it was detailed above, several plant natural substances demonstrate genoprotective effects by complex antioxidant and detoxification activities, neutralisation of carcinogens, and DNA repair promotion. On the other hand, cytotoxic and genotoxic effects of plants and extracts have been demonstrated towards quickly proliferating tumour cells discriminating them against physiologic tissue [197,198]. Below paragraphs illustrate clinically relevant effects applicable to general cancer prevention pre-cancerous lesions-individualised cancer prevention selective cytotoxic effects against malignancies circulating tumour cells and metastatic diseasemitigating strategies phytochemical supplement in optimal palliative care setting as exemplified specifically for colon carcinomas.…”
