Cytotoxic Effects of Tetracycline Analogues (Doxycycline, Minocycline and COL-3) in Acute Myeloid Leukemia HL-60 Cells

Abstract: Tetracycline analogues (TCNAs) have been shown to inhibit matrix metalloproteinases and to induce apoptosis in several cancer cell types. In the present study, the cytotoxic effects of TCNAs doxycycline (DOXY), minocycline (MINO) and chemically modified tetracycline-3 (COL-3) were investigated in the human acute myeloid leukemia HL-60 cell line. Cells were incubated with TCNAs in final concentrations of 0.5–100 µg/ml for 24 h. Viability of the leukemic cells was inhibited in a concentration-dependent manner us… Show more

“…Leukaemia constitutes a heterogeneous group of haematopoietic malignancies and can be categorised in four main types: acute myeloid leukaemia (AML), acute lymphoblastic leukaemia (ALL), chronic myeloid leukaemia (CML) and chronic lymphocytic leukaemia (CLL)1. ALL is referred as the most common paediatric oncological diagnosis23 and overall survival of ALL patients remains relatively poor with 20–40%4.…”

“…IC 50 for bacterial growth was approximately 100 ng/ml while CC 50 was approximately 10 µg/ml, consistent with the CC 50 described previously for doxycycline treatment of the leukocyte lineage, HL-60 cell line. 21 Therefore, overall selectivity determined in this dual-readout experiment was approximately 100.…”

“…The ratio of Bcl-2-to-Bax determines whether a cell survives or undergoes apoptosis [20]. Dox has been shown to both inhibit proliferation and invasiveness of cancer cells [35][36][37], and also to protect neurocytes from apoptosis by directly inhibiting the enzymatic activity of PARP-1 [6,38,39]. However, the details of the mechanism involved remain unclear.…”
