Cytotoxicity and NMR Studies of Platinum Complexes with Cyclooctadiene Ligands

Abstract: The synthesis of a series of platinum complexes containing cyclooctadiene ligands with the general structure PtMeL(R-cod) (where L = Cl, I, nC 3 F 7 , iC 3 F 7 , nC 8 F 17 , Me, aryl, alkynyl and R = H, Me, Et, iPr, nBu, iBu, nHex, Ph) is presented. All complexes are remarkably stable and were obtained in excellent yields. Their structure in both solution and the solid state were explored by crystal structures and multinuclear ( 1 H, 13 C, 19 F, 195 Pt) NMR spectroscopy. Cytotoxicity experiments with selected… Show more

“…correlations from the nitrogen-attached carbonyl to the aromatic protons, whereas the ring carbonyl displays no coupling to 1 H as it is too far away. The 1 H-coupled 13 and increase to 1338 cm -1 respectively, upon coordination to platinum(II) in complex 3.…”

“…Assignment of peaks was assisted by DFT calculations for both N-phenylanthranilic acid and All 1D and 2D NMR spectra ( 1 H, 13 C and 31 P, COSY, NOESY, DEPT, HSQC, HMBC and H2BC) were recorded on a Bruker AVIII-400 spectrometer in CDCl 3 (unless otherwise specified in-text) using BBI or BBFO probes, depending on the nucleus of the sample. Standard Bruker supplied pulse programmes were used for each experiment, with parameters including spectral window, number of increments, pulse angle and repetition rate altered as appropriate for each specific sample.…”

“…While the reaction catalysed with unsaturated polyfluorinated PEPPSI analogue 25 gave moderate 65% yield of the product, 4-methylbiphenyl (28) 62,63 In keeping with standard practice in heavy fluorous chemistry, compound identity and purity were gauged from HRMS data and NMR spectra. [64][65][66][67][68][69][70][71] All reactions were performed in a dry inert atmosphere (Ar) in oven-dried flasks. In the reactions including ruthenium or palladium, solids were introduced into the reaction flasks in a glove box.…”

“…The use of organometallic platinum complexes as potential cytotoxic agents is an area of growing interest, providing an alternative to traditional coordination complexes based on the structural motif of cisplatin. [8][9][10] In this present study, we have expanded upon our initial findings in this area and prepared new organometallic platinum complexes to investigate if these types of compounds are also cytotoxic against other types of cancer, such as breast cancer and Renal Cell Carcinoma (RCC). RCC is the major type of kidney cancer …”
