Cytotoxicity of Ear Drop Excipients in Human and Mouse Tympanic Membrane Fibroblasts

Abstract: Objective Commercial ear drops contain ingredients reported to be inactive. We sought to evaluate such excipients for possible cytotoxicity on human and mouse tympanic membrane (TM) fibroblasts. Study Design Prospective, in vitro. Setting Tertiary academic center. Subjects and Methods Mouse and human TM fibroblasts were treated with 1:10 dilutions of benzalkonium chloride (BKC) 0.0025%, 0.006%, or 0.01%; benzyl alcohol 0.9%; polysorbate 80 (PSB) 2.5%; glycerin 2.4%; povidone 0.2%; or water (control), twice wit… Show more

“…DMSO, as a common cryoprotectant in practical applications, was used to compare with peptoids in a cytotoxicity test. Generally speaking, samples with a cell survival rate of less than 75% are considered to be cytotoxic [36]. The results show that the relative cell viability of all peptoid oligomer solutions was greater than 75% at a concentration of 10 mg/mL, which is considered to be non-cytotoxic (Figure 4).…”

“…Upon matching, the types of medications taken by participants were primarily focused on treating conditions such as diabetes, hypertension, infections, and hyperlipidemia, among others. As detailed information regarding participants' medication, including dosage, duration, administration method, and brand was unavailable, we relied on clinical drug experience and existing literature evidence to primarily define participants taking the following 11 drugs (acetaminophen [21], hydrocodone [22], ciprofloxacin [23], phenytoin [24], levofloxacin [25], rifampin [26], minocycline [27], aspirin [28], metronidazole [29], nitroglycerin [30], and bumetanide [31]) as a high-risk group for drug-induced hearing loss and subsequently excluded them from the study (n = 46).…”
