Cytotoxicity of the Sesquiterpene Lactones, Ivalin and Parthenolide in Murine Muscle Cell Lines and Their Effect on Desmin, a Cytoskeletal Intermediate Filament

Abstract: Vermeersiekte or “vomiting disease” is an economically important disease of ruminants following ingestion of Geigeria (G.) species in South Africa. Sheep are more susceptible, and poisoning is characterized by stiffness, regurgitation, bloat, paresis, and paralysis. Various sesquiterpene lactones have been implicated as the cause of poisoning. The in vitro cytotoxicity of two sesquiterpene lactones, namely, ivalin (purified from Geigeria aspera) and parthenolide (a commercially available sesquiterpene lactone)… Show more

“…Concerning the molecular mechanism underlying cancer cell-induced CAF production, our investigation involved RNA sequencing analysis, revealing DESMIN as the gene exhibiting the highest upregulation in CAFs following lactate treatment. DESMIN, a muscle-specific intermediate filament, plays a vital role in the cytoskeletal framework or scaffold, maintaining cell structure [ 56 ]. Its significance extends to maintaining the structural integrity and organization of muscle tissues, particularly in cardiac and skeletal muscles [ 57 , 58 ].…”
