Czapek-Dox agar containing iprodione and dicloran as a selective medium for the isolation of Fusarium species

Abstract: A new selective medium for Fusarium species has been developed using Czapek‐Dox agar (CZ) containing the fungicides iprodione (3 mg/1) and dicloran (2 mg/1). This new medium (CZID) is selective against numerous species of Alternaria, Epicoccum, Penicillium and mucoraceous fungi. CZID was compared with CZ using samples of barley, malt, sorghum, bean and pea. Fusarium species produced large and easily recognizable colonies on CZID while isolates of Alternaria, Epicoccum and Rhizopus were significantly restricted… Show more

“…Monitoring of fungal viability in soil in treatment 4 was done by taking 1 g samples of soil from a layer 1-2 cm under the soil surface. Samples were diluted in 50 ml SDW, filtered through sterile cheese-cloth, and 500 μl were spread on Czapek-Dox iprodione dichloran (CZID) agar (Abildgren et al 1987). Plates were incubated for 1 week in dark at room temperature, upon which the number of white fungal colonies was counted.…”

“…Kernels of each sample were surface sterilized for two minutes in 1.3% aqueous sodium hypochlorite, rinsed twice with sterile distilled water and transferred onto Czapek-Dox-Iprodion-Dichloran (CZID) agar (Abildgren et al 1987). After incubation at 23°C for 6 days, the percentage of infected kernels was recorded.…”

“…For isolation of Fusarium species, wheat kernels were surface-sterilized using NaOCl (1.3% available chlorine) for 2 min and placed into Petri dishes (5 kernels per dish) containing Czapek-iprodione-dicloran agar (CZID-agar) prepared according to Abildgren et al (1987). After incubation at 22°C for 7 days under near-UV light mycelium growing from the kernels was transferred onto SNA (Nirenberg 1976) and PDA and grown at 22°C for 7 days.…”
