Daidzin inhibits mitochondrial aldehyde dehydrogenase and suppresses ethanol intake of Syrian golden hamsters

Abstract: Daidzin is the major active principle in extracts of radix puerariae, a traditional Chinese medication that suppresses the ethanol intake of Syrian golden hamsters. It is the first isof lavone recognized to have this effect. Daidzin is also a potent and selective inhibitor of human mitochondrial aldehyde dehydrogenase (ALDH-2). To establish a link between these two activities, we have tested a series of synthetic structural analogs of daidzin. The results demonstrate a direct correlation between ALDH-2 inhibit… Show more

“…Presumably, 7,8-dihydroxyflavone might not inhibit aromatase, either. In addition to the activity described above, 7,8-dihydroxyflavone also has been shown to inhibit aldehyde dehydrogenase and estrogen sulfotransferase in vitro with K i values of 35 μM and 1-3 μM, respectively (23,24). However, these actions have not been confirmed in animals.…”

“…In Oriental medicine, medications based on Puerariae radix have been found to be useful in the treatment of alcoholrelated problems, as an anti-intoxication and anti-drinking agent and in the treatment of various liver diseases caused by alcohol abuse (14). In the present study, the effects of Puerariae radix on cell proliferation and NOS expression in the dentate gyrus of rats in the process of growing up *Corresponding author.…”

“…Puerariae radix, one of the most commonly utilized Oriental herbs in the treatment of alcohol-related problems, is known to inhibit mitochondrial aldehyde dehydrogenase and to suppress ethanol intake in Syrian golden hamsters. 1) Puerariae radix is comprised of puerarin, daidzin, daidzein, genistein, and biochanin A; of these, daidzin and daidzein, the major active constituents of Puerariae radix extracts, are known to suppress ethanol intake. 19,20) In a previous study, the inhibitory effect of Puerariae flos on alcohol-induced apoptosis, which appears to act by inhibiting caspase-3 mRNA expression, in neuroblastoma cells was reported.…”
