Introduction. The development of new antifungal drugs has become a global priority, given the increasing cases of fungal diseases together with the rising resistance to available antifungal drugs. In this scenario, drug repositioning has emerged as an alternative for such development, with advantages such as reduced research time and costs. Gap statement. Propafenone is an antiarrhythmic drug whose antifungal activity is poorly described, being a good candidate for further study. Aim. This study aims to evaluate propafenone activity against different species of Candida spp. to evaluate its combination with standard antifungals, as well as its possible action mechanism. Methodology. To this end, we carried out tests against strains of Candida albicans, Candida auris, Candida parapsilosis, Candida tropicalis, Candida glabrata and Candida krusei based on the evaluation of the MIC, minimum fungicidal concentration and tolerance level, along with checkerboard and flow cytometry tests with clinical strains and cell structure analysis by scanning electron microscopy (SEM). Results. The results showed that propafenone has a 50% MIC ranging from 32 to 256 µg ml−1, with fungicidal activity and positive interactions with itraconazole in 83.3% of the strains evaluated. The effects of the treatments observed by SEM were extensive damage to the cell structure, while flow cytometry revealed the apoptotic potential of propafenone against Candida spp. Conclusion. Taken together, these results indicate that propafenone has the potential for repositioning as an antifungal drug.

A modified method of glutaraldeyde-osmium tetroxide fixation was adjusted to characterize the ultrastructure of Candida albicans pleomorphic forms, using phase-contrast microscopy, scanning electron microscopy and transmission electron microscopy. The discovered morphological criteria defining the individual morphotypes are discussed in terms of mycological and histopathological diagnostics of candidiasis. The relations are discussed between fungal pleomorphism, virulence and susceptibility of different morphotypes to fungicides.

Effect of disinfectants on biofilm development by five species of Candida

Candida biofilms have become an increasingly significant clinical problem for hospital-acquired and device-associated infections due to their increased levels of resistance to the antifungals and disinfectants. This study examines the biofilm formation of five Candida isolates (Candida albicans, Candida glabrata, Candida parapsilosis, Candida krusei and Candida tropicalis), using the XTT (2, 3-Bis [2-methoxy-4-nitro-5-(sulfenylamino) carbonyl-2H-tetrazolium-hydroxide]), and compares the susceptibility of biofilm cells to five disinfectants (glutaraldehyde, hydrogen peroxide, peracetic acid, ortho-phtalaldehyde and sodium hypochlorite) at the different times of biofilm formation (6th, 12th, 24th and 48th). Thus, the aim of our study was to investigate whether these disinfectants can interfere on Candida biofilm development phases. Our results indicated that the many variations in biofilm formation in clinical Candida species following disinfectant stress, however, none of the disinfectants completely removed the biofilm. Also, our transmission electron microscopic findings suggest that the disinfectants showed an important effect on the planktonic Candida cell. All these results also emphasize the importance of regular disinfection, before the starting of biofilm formation. On the other hand, XTT assay may not be the only adequate method to evaluate the efficacy of disinfectants. For these reasons, further studies are necessary to evaluate the disinfectant effectiveness on Candida biofilms.
