Danshenol A inhibits TNF-α-induced expression of intercellular adhesion molecule-1 (ICAM-1) mediated by NOX4 in endothelial cells

Abstract: ICAM-1 overexpression and subsequent adhesion of leukocytes to endothelial cells play critical roles in the early stage of atherosclerosis. Danshenol A (DA) is an abietane-type diterpenoid isolated from traditional Chinese herb Salvia miltiorrhiza Bunge. The mechanisms under its regulation of adhesion of molecular expression are explored. Here, the effect of DA on TNF-α-induced ICAM-1 expression was investigated in endothelial cells. TNF-α-induced ICAM-1 expression and subsequent adhesion of monocytes, as well… Show more

“…Additionally, MME is downregulated by NF-κB pathway via microRNA miR155 in B-lymphoma cells [31]. The pro-inflammatory cytokine TNFα activates NF-κB pathway [22], which subsequently upregulates ICAM-1 (intercellular adhesion molecule 1) expression [32]. TNFα is increased in cord blood when mothers are overweight [27], and fpEC also respond to TNFα with an induction of ICAM-1 expression [33].…”

“…Therefore, Danshen represents a traditional Chinese medicine (TCM) that has a relatively high safety profile. To date, the chemical constituents of Danshen have been well identified, including more than 30 lipophilic compounds that have a diterpene chinone structure (tanshinone I-VI, cryptotanshinone, isotanshinone I-II, Danshenol A etc) and more than 50 hydrophilic compounds that mainly have a phenolic acid structure (Danshensu, salvianolic acid A, salvianolic acid B, protocatechuic aldehyde, etc) [1,[5][6][7] . More recently, Tasly Pharmaceuticals, Inc has completed a Phase III clinical trial to evaluate the safety and efficacy of Dantonic® (T89, also known as Compound Danshen Dripping Pills) in patients with chronic stable angina pectoris (ClinicalTrials.gov Identifier: NCT01659580).…”

“…In the endothelial cells, TNFα has been shown to inhibit endothelial nitric oxide synthase (eNOS) expression via the destabilization of eNOS mRNA [9] and increase the expression of the adhesion molecules via the activation of the NFκB pathway [10]. In the endothelial and vascular smooth muscle cells, TNFα activates NADPH oxidase to induce oxidative stress [11].…”
