Darier-White disease in siblings responding to isotretinoin

Abstract: Darier-White disease (keratosis follicularis) is a rare disorder of keratinization involving the epidermis, mucous membranes, and nails. It is said to occur as a result of mutation in the ATP2A2 gene located on chromosome 12q23-24.1. In this article we present the case of two brothers with exacerbations of Darier-White disease who responded very well to systemic retinoids without any side effects within 2 weeks of commencing treatment.

“…Bhat et al used oral isotretinoin at a dose of 20 and 40 mg/day along with a steroid taper for 2 patients with diffuse Darier disease with nail involvement with marked improvement. 37 Alitretinoin (9-cis retinoic acid) was used for a total of 7 patients with rather generalized Darier disease, all of which showed marked improvement of symptoms at a dose of 30 mg/day over a period of at least 1 month; 6 of them, however, relapsed. [38][39][40][41][42] Alitretinoin at 30 mg/day was also used in association with 5-FU on a daily basis in a 43-year-old male who achieved complete remission after 4 months.…”
