DARPin_9-29-Targeted Mini Gold Nanorods Specifically Eliminate HER2-Overexpressing Cancer Cells

Abstract: We have demonstrated that designed ankyrin repeat protein (DARPin) _9-29, which specifically targets human epidermal growth factor receptor 2 (HER2), binds tightly to gold mini nanorods (GNRs). Molecular dynamic simulations showed that a single layer of DARPin_9-29 molecules is formed on the surface of the nanorod and that conjugation with the nanorod does not involve the protein's domain responsible for specific binding to HER2. The nanorod-DARPin (DARPin-GNR) conjugate is specifically bound (in nanomolar con… Show more

“…In animal models, far-red fluorescent proteins, such as mCherry, can be applied to allow intravital visualization of a tumor [38]. Cherry and HER2-specific DARPin 9_29 were fused to obtain the recombinant protein DARPin-mCherry, which specifically stains HER2positive cancer cells [39] and is used for the functionalization of nanoparticles [40][41][42][43] as described below.…”

“…NaYF 4 : Yb 3 + Tm 3 + / NaYF 4 particles emitting ultraviolet radiation when exposed to infrared light were coated with the DARPin-mCherry protein [39], which allows visualization of cancer cells thanks to the far-red fluorescent mCherry module [40]. DARPin 9_29 and the DARPin-mCherry protein containing it were also used to coat 5-nm gold nanoparticles [41] and gold nanorods [42]. DARPin was efficiently coupled with the particle surface to form a crown consisting of approximately 35 protein molecules, thus reducing particle aggregation.…”

“…The IL-37 receptor complex is similar to the IL-18 receptor, another representative of the IL-1 family. IL-18 is one of the key pro-inflammatory cytokines acting as a pathogenetic factor in a number of diseases [41,42]. The IL-18 receptor complex consists of two chains: α (IL-18Rα) and β (IL-18Rβ), each of which has a TIR domain [43].…”

“…In recent years, phototherapy has significantly advanced thanks to the use of lasers as light sources; nano-objects for the delivery of sensitizers [ 10 - 13 ]; targeted dyes [ 14 , 15 ]; increased dye circulation time in the blood [ 16 ]; and sustained release of dyes [ 17 ]. Also, conjugation of dyes with immunoadjuvants is promising in photoimmunotherapy because it leads to the triggering of a systemic immune response [ 18 ].…”
