Daxx-β and Daxx-γ, Two Novel Splice Variants of the Transcriptional Co-repressor Daxx

Abstract: Daxx is involved in transcriptional control and apoptosis. It comprises several domains, including a regulatory C terminus that is responsible for the interaction with numerous proteins such as p53, promyelocytic leukemia protein (PML), and Hsp27. Here, we describe the identification and characterization of two novel variants of Daxx termed Daxx-␤ and Daxx-␥, which are generated by alternative splicing. Alternative splicing results in a truncated regulatory C terminus in both proteins. As a consequence, Daxx-␤… Show more

“…The two novel variants of Daxx were termed Daxx-and Daxx-γ, and these variants are generated by alternative splicing. They have a truncated regulatory C terminus, and Daxx-and Daxx-γ show markedly decreased affinities to PML and have a different nuclear distribution [62] . In summary, all three of the prototypical proteins (PML, SP100, and Daxx) of ND10 share some similar characteristics: (1) They colocalize with ND10.…”

“…For the Rb proteins, these clearly represent the hypophosphorylated forms of the proteins. For Daxx, it is presently unclear whether faster migrating bands represent different phosphoisoforms or splice variants [89] of the protein.…”

“…38, 39 Several splice variants of Daxx that differ at the C terminus and with regard to their ability to interact with PML have been described. 40 Daxx is a well-established regulator of transcription. Daxx binds to the transcriptional coregulators, CREB-binding protein and histone deacetylase, to DNA methyltransferases 41 as well as to numerous transcription factors, including members of the Pax and p53 families, C/EBPβ, ETS1, SMAD4 and glucocorticoid and androgen receptors.…”
