dbEST‐derived microsatellite markers in celery (Apium graveolens L. var. dulce)

Abstract: We report on the development of 11 (from database expressed sequence tags) dbEST-derived microsatellite markers in celery ( Apium graveolens L. var. dulce ). The sequences were obtained from DNA accessions available from GenBank and contained di-, tri-and pentanucleotidic motifs. All the microsatellites were found in expressed sequence tags and they are expected to become useful tools for ecological, genetic and evolutionary studies, as well as for celery breeding. Polymorphism was explored in 16 celery commer… Show more

“…The SSR sequences were screened using Cap 3 software (Huang and Madan 1999) and the following parameters were adopted: at least eight repeats for di-, six repeats for tri-and tetra-, and three repeats for penta-nucleotidic (Acquadro et al 2006 (20) and Psetta maxima (28)] were selected randomly for polymerase chain reaction Conserv Genet (2009) 10: [1629][1630][1631] (PCR) in L. polyactis. Primers were designed using the software Primer 5.0 adopting default parameters.…”

“…The SSRs were obtained by screening through a total of 10,570 ESTs of Argopecten irradians and Chlamys farreri available from GenBank (http://www.ncbi.nlm.nih.gov/ Genbank/). The SSR sequences were screened using Cap 3 software (Huang and Madan 1999) and the following parameters were adopted: at least eight repeats for di-, five repeats for tri-and tetra-, and three repeats for pentanucleotidic (Acquadro et al 2006). Overall, 81 different EST sequences containing SSRs were derived and then 20 ESTSSRs were selected randomly for polymerase chain reaction (PCR) in P. yessoensis.…”
