DDR2 Expression in Cancer-Associated Fibroblasts Promotes Ovarian Cancer Tumor Invasion and Metastasis through Periostin-ITGB1

Abstract: Ovarian cancer has the highest mortality of all gynecologic malignancies. As such, there is a need to identify molecular mechanisms that underlie tumor metastasis in ovarian cancer. Increased expression of receptor tyrosine kinase, DDR2, has been associated with worse patient survival. Identifying downstream targets of DDR2 may allow specific modulation of ovarian cancer metastatic pathways. Additionally, stromal cells play a critical role in metastasis. The crosstalk between tumor and stromal cells can lead t… Show more

“…Media secreted by Ddr2-depleted human ovarian tumor CAFs leads to decreased ovarian tumor cell invasion and migration compared to media secreted by Ddr2-expressing CAFs [ 25 ]. We confirmed this result in Matrigel invasion assays in Boyden chambers using two human ovarian tumor cell lines (Tyknu; OVCAR8) and CAF conditioned media (CM) added to the lower well (Fig.…”

“…Fascia was significantly associated with the ITGB1 expression, and their co-expression was significantly associated with shorter disease-free and overall survival rate [49]. Lung metastasis and the migration of glioblastoma and osteosarcoma cells were also facilitated by the association between α9β1 integrin and TN-C [87].…”

“…Meanwhile, integrins can also be expressed in CAFs to affect tumor cells. In OC, CAFs increase the expression level of ITGB1 through DDR2, further increasing the expression of POSTN in CAFs, which in turn promotes the development of invasive metastasis in OC cells [87]. In CRC, PNs in CAFs can lead to phosphorylation of AKT via α5β1 or α6β4 integrins affecting autophagymediated EMT and invasive metastasis in CRC cells [88].…”
