“…In order to estimate the optical distortion occurring in our preparations, empirical point spread functions (PSF) were generated by collecting light at different depths (in 0.8 μm steps) from fluorescent beads (0.04-μm diameter) injected into the granular and molecular layer in sagittal and coronal cerebellar slices (Yae et al, 1992) and processing signals off-line (Image-J). In the granular layer, which is almost isotropic, PSF analysis showed that light signals vanished in ∼10 μm both vertically and horizontally.…”

“…In searching for less phototoxic 180 A.L. Obaid et al / Journal of Neuroscience Methods 134 (2004) 179-190 and more soluble dyes with large voltage-sensitive fluorescence signals, we first assayed the dienylstyryl-pyridinium dye RH795, originally synthesized by Grinvald et al (1994) and now available from molecular probes (R649), which is highly water soluble and has already found numerous applications in voltage-sensitive dye recordings of electrical activity (Albowitz and Kuhnt, 1993;Contreras and Llinas, 2001;Maeda et al, 2001;Yae et al, 1992). Because we found that RH795 is readily internalized by the neurons of the submucous plexus, we attempted to generate chimeric structures that retained the best features of both of these dyes.…”
