Decellularization protocols of porcine heart valves differ importantly in efficiency of cell removal and susceptibility of the matrix to recellularization with human vascular cells

Abstract: Compared with trypsin and sodium-dodecyl-sulphate involving decellularization procedures, reported to be effective in cell removal and susceptible to recellularization with human cells, only the porcine matrix treated with a new detergent-based decellularization method using 0.25% tert-octylphenyl-polyoxyethylen/sodium-deoxycholate followed by nuclease digestion presented an excellent scaffold for recellularization with human cells.

“…Many organs, e.g. urinary bladder [6], small intestinal [7][8][9][10][11], heart valves [12][13][14][15], skin [16,17], liver [18], anterior crucial ligament bone [19], kidney [20], pericardium [21], blood vessels [22], nerves [23], skeletal muscle [24], tendon [25] and vocal cord [26,27] from bovine and porcine tissues, have been used for fabricating the ECMbased scaffold. But, the use of goat-lung has not been focused for tissue engineering applications.…”

“…Decellularization with 0.25% Triton X-100 (Fischer Scientific, Pittsburgh, PA) and 0.25% deoxycholic acid (DOC; Alfa Aesar Ward Hill, MA) was performed using a modification of the methods reported by Rieder et al 13 Briefly, lymph nodes were digested in a solution of 0.25% Triton X/ 0.25% DOC for 24 h at 37°C. Tissues were subsequently washed in sterile PBS at 4°C for 24 h with serial changes of PBS.…”
