The endemic and critically endangered cyprinid Chondrostoma lusitanicum has a very restricted distribution range. In order to estimate genetic diversity, characterize population structure and infer the demographic history, we examined six microsatellite loci and cytochrome b (mtDNA) sequences from samples taken throughout C. lusitanicum's geographical range. Estimates of genetic diversity were low in all samples (average He \ 0.35). The microsatellite data pointed to a major difference between northern (Samarra and Tejo drainages) and southern (Sado and Sines drainages) samples. This separation was not so clear with mtDNA, since one sample from the Tejo drainage grouped with the southern samples. This could be related with ancestral polymorphism or with admixture events between northern and southern sites during the late Pleistocene. Nevertheless, both markers indicate high levels of population differentiation in the north (for microsatellites F ST [ 0.23; and for mtDNA U ST [ 0.74) and lower levels in the south (F ST \ 0.05; U ST \ 0.40). With microsatellites we detected strong signals of a recent population decrease in effective size, by more than one order of magnitude, starting in the last centuries. This is consistent with field observations reporting a severe anthropogenic-driven population decline in the last decades. On the contrary mtDNA suggested a much older expansion. Overall, these results suggest that the distribution of genetic diversity in C. lusitanicum is the result of both ancient events related with drainage system formation, and recent human activities. The potential effect of population substructure generating genetic patterns similar to a population decrease is discussed, as well as the implications of these results for the conservation of C. lusitanicum.

Characterization, management and protection of biodiversity are the most prominent challenges in conservation biology. Analyses on molecular similarity (Random Amplified Polymorphic DNA-Polymerase Chain Reaction, RAPD-PCR) and morphologic patterns (morphometric and meristic characters) were performed to evaluate the existence of population structuring in the Doce river basin, in a widespread small-sized characin, Astyanax bimaculatus. Three hundred (300) individuals were collected from six locations isolated by waterfalls or dams, in the Doce river basin, Minas Gerais State, Brazil. Genetic differentiation among tributaries was significant (p<0.00001) and accounted for 21% of total variance (p<0.00001). Only the Grande waterfall of the Casca river was an effective factor for differentiation of populations (p<0.00001), and meristic characters were also consistent with molecular data. We concluded that genetic and morphologic variation of this species was not necessarily associated with waterfalls and that molecular and meristic data are effective predictors of population divergence in this basin. The consequences of these findings for the protection and management of aquatic biodiversity were discussed.

The phylogeographic structure of vairone (Telestes muticellus), a primary freshwater fish endangered in a large part of its distributional range, was assessed: i) to reconstruct the complex dispersion pattern in the upper Tyrrhenian hydrographic basins of Ligury, actually not recognised as peri-Mediterranean ichthyogeographic district, and ii) to evidence the shape of population genetic structure as useful tool for future conservation strategies. A partial fragment of mitochondrial DNA cytochrome b (497 bp) was sequenced in 109 specimens sampled from 8 populations, along an east-west geographic gradient. Fourteen haplotypes were identified, confirming the evolutionary distance between the two co-generic species: T. muticellus as ‘Ligurian’ clade and T. souffia as ‘French’ clade. The Nested Clade Analysis (NCA), the population genetic variability and population structure suggested a natural colonization occurred throughout the crossing of Alpine and Apennine watershed. The hierarchical Analysis of Molecular Variance (AMOVA) confirmed a geographic distinction between the populations from western (WTL) and from eastern (ETL) Tyrrhenian basins of Ligury colonised through the river capture processes along the Maritime Alpine watershed (Padano-Venetian district) and along the Apennine watershed (Tuscano-Latium district), respectively.
