Deconvolution improves the detection and quantification of spike transmission gain from spike trains

Abstract: Accurate detection and quantification of spike transmission between neurons is essential for determining neural network mechanisms that govern cognitive functions. Using point process and conductance-based simulations, we found that existing methods for determining neuronal connectivity from spike times are highly affected by burst spiking activity, resulting in over- or underestimation of spike transmission. To improve performance, we developed a mathematical framework for decomposing the cross-correlation be… Show more

“…To further examine the extent to which washing out gabazine returned endogenous inhibitory activity as GABA A receptors are released from blockade, we also computed the spike transmission probability (STP; Supplementary Figure 13 ). This cross-correlogram based metric has been used to infer putative inhibitory functional connectivity from ongoing extracellular spiking activity and to identify neuron pairs with a reduction in spike transmission gain (STG) 102 . Indeed, we find that washout of gabazine led to a significant increase in the number of putative inhibitory connections (Wilcoxon signed rank: p=0.012; Figure 6b ).…”

“…To this end, putative monosynaptic excitatory (PMSE) pairs were sought between all possible mPFC and DMS pairs (Fig.7A). PMSE pairs were identified based on well-established methods with minor modifications 33,34 . The criterion is based on a strongly increased likelihood of spiking of the postsynaptic pair member with minimal jitter within a narrow time window (monosynaptic delay) after the presynaptic member’s spikes (see methods for details).…”

“…To determine whether a monosynaptic connection may exist between units, count cross-correlation histograms (CCHs; 0.1 ms bins) were constructed for putative pre- and postsynaptic spike train pairs as previously described 93 . Briefly, the spike transmission curve was estimated by the difference between the deconvolved CCH and the baseline, determined by hollowed median filtering of the count CCH, scaled to spikes/s.…”
