Decreased Expression of the Epithelial Ca 2+ Channel TRPV5 and TRPV6 in Human Renal Cell Carcinoma Associated With Vitamin D Receptor

Abstract: Decreased TRPV5/V6 expression was noted in renal cell carcinoma, which correlated with vitamin D receptor. Different expression was also detected among the different renal cell carcinoma histopathological subtypes. Our observations suggest that altered vitamin D receptor expression may be associated with renal cell carcinoma carcinogenesis via TRPV5/6.

“…The TRPV6 channel is a calcium cation channel protein, which is the major constituent of superfamily of transient receptor potential (TRP) channels, further in terms of subfamily vanilloid member 6, which regulates the calcium homeostasis in the epithelial tissue of human organs (3). TRPV6 is overexpressed in colon cancer, breast cancer, prostate cancer, parathyroid cancer and thyroid cancer (3)(4)(5)(6), however decreased TRPV6 expression was observed in esophageal cancer, non-small cell lung cancer and renal cancer (7)(8)(9). Overexpression of TPPV6 in many cancers indicates the idea of this protein as encoded by a probable oncogene, but the level of TRPV6 in other cancers showed a tendency of a tumor-suppressor.…”

“…Cav1.3 is dominantly active during cell depolarization and when calcium and other nutrients such as glucose are plentiful in the diet. In contrast, TRPV6, found in a large concentration in the duodenum, actively recovers calcium as luminal nutrients decrease during cell hyperpolarization and is upregulated by 1,25(OH) 2 D 3 [24, 26, 27]. …”

“…Furthermore, as previously mentioned, calcium absorption is preferentially mediated by TRPV6 when luminal nutrients decrease and TRPV6 is upregulated by 1,25(OH) 2 D 3 [24, 26, 27]. Therefore, in children with GGM, calcium may be preferentially mediated by TRPV6 instead of Cav1.3 in an effort to maximize the uptake of calcium in the context of malabsorption.…”

“…Unlike the other four members of the TRPV family that are non‐selective cation channels, TRPV5 together with TRPV6 is a remarkable Ca 2+ ‐selective channel that serves for apical calcium entry in absorptive and secretory tissues (Nijenhuis et al 2003). However no data have been provided so far regarding the potential involvement of TRPV5 or TRPV6 channels in renal cancers, although the decreased TRPV5/TRPV6 expression was already reported in renal cell carcinoma, which correlated with the vitamin D receptor (Wu et al 2011). On the other hand, endogenous TRPV5 mRNA expression was observed using RT‐PCR analysis in human Jurkat T leukaemia cell line (Vasil’eva et al 2008) and in K562 erythroleukaemia cell line (Semenova et al 2009).…”
