Decreased Platelet 3H-Paroxetine Binding in Obsessive-Compulsive Patients

Abstract: The similarities between the serotonin (5-HT) transporter in both human platelets and human brain permit us to investigate this structure in patients with different psychiatric disorders. Several reports have shown abnormalities of the 5-HT transporter, by means of the measurement of the 5-HT uptake or of the 3H-imipramine binding, in platelets of patients with obsessive-compulsive disorder (OCD). The availability of the ligand 3H-paroxetine, a selective 5-HT reuptake inhibitor, to label … Show more

“…From a conceptual point of view, the presence of the 5-HT transporter in nucleate cells such as lymphocytes, might permit molecular genetic studies in patients, such as those affected by mood and anxiety disorders, where disturbances at the level of the platelet 5-HT transporter have been reported (Meltzer and Lowy 1987;Marazziti et al 1996), although it is possible that the regulation of the 5-HT transporter may be different in nucleated and nonnucleated cells. Pursuing this line of research, recently Lesch et al (1996) showed that a variant of the polymorphism in the 5-HT transporter reduces the expression of the transporter itself and the 5-HT uptake in lymphoblasts.…”

“…Par binding was carried out according to the method of Marazziti et al [19]. The incubation mixture consisted of 100 Ìl of platelet membranes (50-100 Ìg protein/tube), 50 Ìl of 3 H-Par at six concentrations ranging from 0.01 to 1 nM, and 1,850 Ìl of assay buffer.…”

“…Nineteen patients with OCD, out of the total of 21, 20 patients with depression and all healthy controls underwent blood sampling for the evaluation of 3 H-Par binding which was carried out according to the method of Marazziti et al [33]. Briefly, 20 ml of venous blood was drawn from fasting subjects between 8 and 9 a.m., in the months April-June to avoid the possible interference of either circadian or seasonal rhythms, and gently mixed with 1 ml of anticoagulant 0.15 M EDTA.…”
