Deep sequencing of voodoo lily (Amorphophallus konjac): an approach to identify relevant genes involved in the synthesis of the hemicellulose glucomannan

Abstract: A Roche 454 cDNA deep sequencing experiment was performed on a developing corm of Amorphophallus konjac—also known as voodoo lily. The dominant storage polymer in the corm of this plant is the polysaccharide glucomannan, a hemicellulose known to exist in the cell walls of higher plants and a major component of plant biomass derived from softwoods. A total of 246 mega base pairs of sequence data was obtained from which 4,513 distinct contigs were assembled. Within this voodoo lily expressed sequence tag collect… Show more

“…HM is detected in most Arabidopsis cell types (Handford et al, 2003) and facilitates embryogenesis (Goubet et al, 2009), germination (Rodríguez-Gacio et al, 2012), tip growth (Bernal et al, 2008), and vascular development (Benová-Kákosová et al, 2006;Yin et al, 2011). In the last 10 years, in vitro mannan synthase activity has been demonstrated for recombinant CSLA proteins from many land plants (Liepman et al, 2005Suzuki et al, 2006;Gille et al, 2011;Wang et al, 2012). HM synthesis may also involve CELLULOSE SYNTHASE-LIKE D (CSLD) enzymes and MANNAN SYNTHESIS-RELATED (MSR) accessory proteins (Yin et al, 2011;Wang et al, 2013), but their precise roles in relation to the CSLAs have not been established.…”

“…Many of the wall polymers contain O-acetyl substituents, including the hemicelluloses xyloglucan (XyG) (Kiefer et al, 1989), (glucurono-)arabinoxylans (Carpita, 1996;Scheller and Ulvskov, 2010), and (galacto-)glucomannans, which occur in the secondary walls of gymnosperms (Lundqvist et al, 2002) and function as storage polymers (Gille et al, 2011), as well as various pectic polysaccharides (Caffall and Mohnen, 2009) and the polyphenol lignin (Del Río et al, 2007).…”
