Defects in memory B-cell and plasma cell subsets expressing different immunoglobulin-subclasses in patients with CVID and immunoglobulin subclass deficiencies

Abstract: IgAdef, B-cell defects were mainly restricted to surface membrane (sm)IgA 1 PCs and MBCs, with 2 clear subgroups showing strongly decreased numbers of smIgA 1 PCs with mild versus severe smIgA 1 MBC defects and higher frequencies of nonrespiratory tract infections, autoimmunity, and affected family members. “…The importance of using age-matched control subjects is emphasized in the article because many diagnostic laboratories still use adult levels of B-cell subpopulations for comparisons with their patients. Overall decreased IgA 1 1 and/or IgA 2 1 plasma cell counts were found in all patients, except for 2 patients with SIgAD who showed abnormally reduced numbers of surface membrane IgA 1 1 and IgA 2 1 memory B cells, confirming the failure of IgA production, which is in line with severely reduced serum IgA levels.
