Defects in Pulmonary Vasculature and Perinatal Lung Hemorrhage in Mice Heterozygous Null for the Forkhead Box f1 Transcription Factor

Abstract: Decreased pulmonary expression of Forkhead Box f1 (Foxf1) transcription factor was associated with lethal alveolar hemorrhage in 55% of the Foxf1 +/- newborn mice. The severity of the pulmonary abnormalities correlates with the levels of Foxf1 mRNA. Defects in alveolarization and vasculogenesis were observed in subsets of the Foxf1 +/- mice with relatively low levels of expression from the normal Foxf1 allele. Lung hemorrhage was coincident with disruption of the mesenchymal-epithelial cell interfaces in the a… Show more

“…Cell proliferation rates were detected by immunohistochemical staining of mouse lung paraffin sections using mouse monoclonal antibodies specific for BrdU (1:100; clone Bu 20A; Dako, Carpinteria, CA, USA) or PCNA (1:1000; clone PC-10; Roche Diagnostics, Indianapolis, IN, USA). Antibody-antigen complexes were detected by anti-mouse antibody conjugated with alkaline phosphatase and BCIP/NBT substrate (all from Vector Lab) as described (Kalinichenko et al, 2001aKim et al, 2005a). In each mouse lung, we counted the number of BrdU-positive cells per 1000 cells in ten distinct 400 Â microscope fields.…”

“…Nuclear protein extracts were prepared from HL-1 cells, and then subjected to Western Blot analysis (Kalinichenko et al, 2001a) using mouse monoclonal antibody against cyclin B1 (1:500; GNS-11; BD Biosciences), p27 Kip1 (1:3,000; G173-524; BD Biosciences), or p21 Cip1 (1:200; SXM30; BD Biosciences). We also used rabbit polyclonal antibody against Cdk2 (1:1,500; M2; Santa Cruz) for loading controls.…”

“…Foxf1 and Foxf2 are expressed in lung mesenchyme. In addition, Foxf1 is abundant in subepithelial areas and in alveolar endothelial cells, where it regulates mesechyme-epithelial interactions, alveogenesis, and vascular remodelling (Hellqvist et al, 1996;Aitola et al, 2000;Costa et al, 2001;Kalinichenko et al, 2001;Mahlapuu et al, 2001;Lim et al, 2002;Ramirez et al, 2003).…”
