Deletion of GPR30 Drives the Activation of Mitochondrial Uncoupling Respiration to Induce Adipose Thermogenesis in Female Mice

Abstract: Thermogenic adipocytes possess a promising approach to combat obesity with its capability promoting energy metabolism. We previously discovered that deletion of GPR30 (GPRKO), a presumably membrane-associated estrogen receptor, protected female mice from developing obesity, glucose intolerance, and insulin resistance when challenged with a high-fat diet (HFD). In vivo, the metabolic phenotype of wild type (WT) and GPRKO female mice were measured weekly. Acute cold tolerance test was performed. Ex vivo, mitocho… Show more

“…When the UCP-1 protein in BAT is activated, the energy in the body is converted into heat, which declined energy accumulation in the body and inhibits obesity. [37,38] At present, carnosine has been reported to inhibit obesity in rats by activating the expression of the UCP-1 protein in brown adipose tissue. [39] However, anserine and carnosine have similar structures and differ by one methyl group.…”

“…Bone morphogenetic protein 7 (BMP7) has also been shown to upregulate UCP1 and increase adipocyte thermogenesis (26). The absence of the membrane-associated estrogen receptor G proteincoupled receptor 30 (GPR30) may promote BAT mitochondrial uncoupling of respiration (27), suggesting that it is a negative regulator of thermogenesis and contributes to reduced obesity. Optic atrophy protein 1 (OPA1) (28), Brain-derived neurotrophic factor (BDNF) (29), also plays an important role in the adaptive thermogenesis of BAT.…”
