Deletion of the Heptosyltransferase Genes rfaC and rfaF in Escherichia Coli K‐12 Results in an Re‐Type Lipopolysaccharide with a High Degree of 2‐Aminoethanol Phosphate Substitution

Abstract: The chromosomal genes rfaC and rfaF of Escherichia coli W3110 were inactivated by allelic-replacement mutagenesis to generate a defined strain lacking both heptosyltransferases which catalyze in lipopolysaccharide (LPS) biosynthesis the transfer of the first two L-glycero-D-manno-heptose (Hep) residues to 3-deoxy-~-mannm-~-octu~oson~c acid (Kdo). The LPS of the mutant was isolated and its chemical structure was investigated by compositional analysis and nuclear magnetic resonance spectroscopy of isolated, deac… Show more

“…We also found minor peaks (m/z 1,921-1,940; Fig. 3C and data not shown) indicative of additional modifications that could correspond to phosphoethanolamine-or aminodeoxypentose-modified lipid A, molecules that appear when the OM is damaged (39,47). Therefore, even under inducing conditions, our depletion strains have slight defects in LPS biogenesis.…”

“…Many pathogens, such as strains of Pseudomonas aeruginosa in cystic fibrosis patients (29,30), now are resistant to commercially available antibiotics. Therefore it is necessary to develop new antibacterial agents that target previously unexploited systems, such as the enzymes that assemble the lipid A component of lipopolysaccharide (2,8,9,(31)(32)(33)(34)(35). The cytosolic acyltransferase LpxA catalyzes the first step of the lipid A pathway ( Fig.…”

“…Isolation of Kdo 2 -Lipid A-WBB06 cells (27) were grown overnight at 37°C to A 600 ϳ3.5 in 2 liters of LB broth adjusted to pH 7.0 and supplemented with 12 g/ml tetracycline. Cells were harvested by centrifugation at 6,000 ϫ g for 10 min, washed with an equal volume of phosphate-buffered saline (28), and resuspended in 160 ml of phosphate-buffered saline.…”
