Delphinidin prevents high glucose-induced cell proliferation and collagen synthesis by inhibition of NOX-1 and mitochondrial superoxide in mesangial cells

Abstract: This study examined the effect of delphinidin on high glucose-induced cell proliferation and collagen synthesis in mesangial cells. Glucose dose-dependently (5.6-25 mM) increased cell proliferation and collagen I and IV mRNA levels, whereas pretreatment with delphinidin (50 μM) prevented cell proliferation and the increased collagen mRNA levels induced by high glucose (25 mM). High glucose increased reactive oxygen species (ROS) generation, and this was suppressed by pretreating delphinidin or the antioxidant … Show more

“…HG was recognized not only as the main characteristic of DN but also as a risk factor for the occurrence and development of DN [18]. Research con rmed HG can promote proliferation of HMCs [19][20][21]. Our study provides evidence that TRL can inhibit HG-induced HMC overproliferation in a concentration-dependent manner.…”

“…The delphinidin (delph) aglycone has also been studied for its antioxidant properties in other tissue models of diabetes [98]. In a cellular model of diabetic nephropathy, 24 h incubation of mesangial (glomerular) cells with 25 mM glucose induces cellular proliferation and excess extracellular matrix, which are triggered by mitochondrial stress stemming from NADPH oxidase 1 (NOX1) activation and ROS generation.…”

“…The delphinidins evidenced, in previous studies, several bioactive properties, being the anthocyanin compound with the greatest beneficial effect in preventing cancer, being related to the death of apoptotic or autophagic cells in various types of cancer. In addition, it has antioxidant and anti-inflammatory action [58,59].…”
