Deltonin induces apoptosis in MDA-MB-231 human breast cancer cells via reactive oxygen species-mediated mitochondrial dysfunction and ERK/AKT signaling pathways

Abstract: Abstract. Deltonin, a steroidal saponin isolated from Dioscorea zingiberensis Wright, exhibits high cytotoxic activity in cancer cells. In the present study, the effects of deltonin on cell proliferation and apoptosis were evaluated in the MDA-MB-231 human breast carcinoma cell line. Following treatment with deltonin, the viability of MDA-MB-231 cells was analyzed using MTT assay and apoptosis, mitochondrial membrane potential (∆Ψm) alternation and intracellular reactive oxygen species (ROS) generation was det… Show more

“…It is known that the cytotoxic effects of radiation are mediated primarily through increased formation of hydroxyl radicals and ROS that, in turn, can damage proteins and trigger apoptosis in a variety of cells (Morales et al , 1998; Van Laethem et al , 2006; Na et al , 2012; Soga et al , 2012; Zhu et al , 2012; Zhang et al , 2012; Sinha et al , 2013; Zhang et al , 2013). Therefore, to investigate whether the observed IR-induced GI inhibition, paralleled by induction of apoptosis, was associated with ROS formation, the levels of intracellular H 2 O 2 were initially measured with DCF staining.…”

“…Cleaved caspase-3 and cleaved caspase-8 expression have been shown to relate with the mitochondria-dependent apoptotic pathway (Zhang et al, 2013 ; Shih et al, 2017 ). In the present study, the activation of caspase-3 and caspase-8 induced by MPP + could be prevented by the pretreatment of Als.…”

“…Compared with numerous specific inhibitors, which have a single target for the treatment of leukemia, COR may be more effective, as drug resistance frequently emerges due to hyperactivation of alternative signaling pathways under the treatment with a drug that has a single target. As Akt, ERK and caspase-3 signaling are all interconnected and are not separate pathway entities ( 37 ) and COR exhibited characteristics of a multi-targeted therapeutic, it may be deduced that it rarely causes resistance in AMoL, that its efficacy may be higher and that the response duration may be longer.…”
