Demonstration of the Rat Lymphatic Vessels Using Immuohistochemistry and In Situ Hybridization for 5^|^apos;-Nucleotidase

Abstract: The cellular distribution of 5'-nucleotidase (5'-Nase) was studied in the rat tissues at the levels of protein and mRNA by use of immunohistochemistry and in sift: hybridization, and compared with that of reaction products by enzyme-itistochemistiy. 5'-Nase activity, immunoreactivity of ecto-5'-Nase (CD73) and hybridization signals for its 1nRNA were colocalized in the lymphatic vessels including central lacteals of the small intestine, suggesting that 5'-Nase is actually produced in the lymphatic endothelial … Show more

“…This coincides with the results that the regrowing lymphatics in the lesion displayed an accumulation of 5'-Nase transcripts. These findings suggest that the 5'-Nase may be correlated to the functional maturation of the regenerating lymphatics, because 5'-Nase is considered to facilitate membrane transport of lymph (Shimoda et al 2003). “…5'-Nase mRNA was preferentially expressed in the lymphatic vessels in the intestinal muscle coat, as in central lacteals (Shimoda et al 2003). Few or no signals were seen in any other tissue elements including blood vessels.…”

“…Although many of the mice lacking Ang2 die within 2 weeks of birth (Gale et al 2002), we have obtained multiple surviving adult mutant animals with severe lymphatic dysfunction (Hunter et al 2006) in order to examine the fine structure of their lymphatic capillaries. Hence, we have explored the precise structure of the lymphatic capillaries in Ang2 knockout and Ang1 knockin mice by focusing on the lymphatics in the digestive organs, which contain a welldeveloped lymphatic vasculature (Shimoda 1998;Shimoda and Kato 2006;Shimoda et al 1997Shimoda et al , 2001Shimoda et al , 2003Shimoda et al , 2004.…”

“…But the cumulative recovery in tumor-bearing mice was slightly higher than that in healthy mice, indicating enhanced lymphatic uptake in tumor-bearing mice because the tumorinduced lymphangiogenesis (Shimoda et al, 2003) promotes the lymphatic uptake (Sleeman et al, 2009).…”
