Dengue Virus Non-Structural Protein 5

Abstract: The World Health Organization estimates that the yearly number of dengue cases averages 390 million. This mosquito-borne virus disease is endemic in over 100 countries and will probably continue spreading, given the observed trend in global warming. So far, there is no antiviral drug available against dengue, but a vaccine has been recently marketed. Dengue virus also serves as a prototype for the study of other pathogenic flaviviruses that are emerging, like West Nile virus and Zika virus. Upon viral entry in… Show more

“…Dengue virus (DENV), a mosquitoborne, positive-strand RNA virus from the family Flaviviridae, has emerged as one of the major public health concerns in India and recent estimates suggest that over 60 million people globally get infected with DENV every year (10). The crystal structures of NS5 protein of DENV and West Nile virus have identified zinc binding site in RdRp domain and propose an important structural role for zinc ions in polymerase activity (11,12). Therefore, zinc is likely to play a prominent role in the polymerase activity of DENV.…”

“…Among all the proteins of dengue virus, RNA-dependent RNA polymerase (RdRp) is encoded by non-structural gene NS5 and consists of 20 important enzymatic activities needed for viral propagation. Studies have been conducted on RdRp for designing drug against dengue virus as it is known to be the largest and highly conserved protein (Noble et al, 2010;Noble et al, 2013;El Sahili and Lescar, 2017;Lim et al, 2015). Conserved motif 'YQNKVVRVQRPAKNGTVM-DVISSRDQRGSGQVGTYGLNTFTNMEAQLIRQ' was found in NS5 ( Fig.…”

“…The E protein has been implicated in the membrane fusion between host cells and DENV particles [11]. Importantly, non-structural proteins regulate various stages of the DENV lifecycle, including viral RNA replication, virion assembly, polyprotein cleavage, maturation, and defense against host immunity [12][13][14][15]. Antiviral development approaches thus far have targeted both structural and non-structural proteins, with the main focus on multifunctional enzymes such as NS3, NS4B and NS5 [16][17][18][19][20].…”
