Deoxygenation-Induced Non-Electrolyte Pathway in Red Cells from Sickle Cell Patients

Abstract: Red cells from patients with sickle cell disease contain HbS rather than the normal HbA (here termed HbS cells). On deoxygenation, HbS cells exhibit a distinctive solute permeability pathway, Psickle, activated stochastically, and partially inhibited by DIDS and dipyridamole. It is often referred to as a cation channel although its permeability characteristics remain vague and its molecular identity is unknown. We show that, in contrast to normal red cells, a proportion of HbS cells underwent haemol… Show more

“…Eryptosis participates in the pathophysiology of several clinical conditions, including sepsis [119], hemolytic uremic syndrome [120], renal insuficiency [121], malaria [10], sickle-cell anemia [122,123], beta-thalassemia [80], glucose-6-phosphate dehydrogenase (G6PD)-deficiency [80], phosphate depletion [124], and Wilson's disease [118]. Enhanced eryptosis in sepsis [119] and hemolytic uremic syndrome [120] is secondary to the capability of serum from respective patients to trigger eryptosis.…”

“…P sickle has the characteristics of a nonspecific cation channel [34]. An anion permeability is controversial, whilst, more recently, it has been proposed as permeable under certain conditions to nonelectrolytes [49]. The main effect of P sickle is probably the increased Ca 2+ entry [49, 50] and possibly the Mg 2+ loss [45].…”

The main effect of P sickle is probably the increased Ca 2+ entry [49, 50] and possibly the Mg 2+ loss [45]. Raised intracellular Ca 2+ has several roles which include phospholipid scrambling [51].…”

“…The present short review compiles the present knowledge on mechanisms regulating, triggering and inhibiting the suicidal erythrocyte death or eryptosis. For a more detailed discussion of mechanisms involved, the reader is referred to earlier, more extensive reviews [5][6][7][11][12][13].…”
