Deoxyribonucleic acid polymerase associated with feline leukemia and sarcoma viruses: Properties of the enzyme and its product

Abstract: Native caw thymus and salmon sperm D N A are better templates than denatured calf thymus and salmon sperm D N A for the DNA-dependent D N A polymerase activity of feline sarcoma virus. This enzyme activity of feline sarcoma virus and feline leukemia virus faithfully replicates dA T copolymer to synthesize a sequence of alternate deoxyadenylate-deoxythymidylate. With a homopolymeric duplex, polydA.dT, the DNAdirected enzyme from feline sarcoma virus or feline leukemia virus indicates a requirement for simultane… Show more

“…I Reaction mixtures (0.2 ml) were cooled at 0 to 4 C after treatment at specified conditions; 100 ;tg (0.1 ml) of bovine serum albumin was added, and the proteins were precipitated by addition of 0.2 ml of cold 50% trichloroacetic acid. The acid-insoluble materials were collected on membrane filters (Millipore Corp.), washed, dried, and counted for radioactivity as described previously (21…”

“…cells were grown in 50% phosphate-free minimal essential medium containing 10% dialyzed fetal bovine serum in the presence of the 3H-labeled L-amino acid mixture (2 gCi/ml) and carrier-free [32P]phosphate (80 ACi/ml). Two changes of the medium were made at 24-h intervals, and the virus was purified from the pooled culture fluids by the published procedure (19,21).…”

“…Two changes of the medium were made at 24-h intervals and the virus was purified from the pooled culture fluids by the published procedure (19,22). The pelleted virus was then disrupted in 20 mM Tris-hydrochloride (pH 7.4) containing 0.1% Nonidet P-40 at 0 C for 1 h and then treated with RNase (0.5 mg/ml, final concentration) at 37 C for 2 h. The solution was then brought to 8 M Gu .HCI and 0.3% ,8-mercaptoethanol, heated at 56 C for 45 min and applied on a Bio Gel A 5-m column (1.5 by 87 cm) equilibrated with 6 M Gu-HCl for fractionation of virion polypeptides (15,19).…”

“…Viruses were harvested from 24-h culture fluids of virus-infected confluent cells for complementary DNA (cDNA) synthesis and from 6-to 8-h culture fluids for polyadenylic acid [poly(A)]-containing virus RNA. Virus was purified from culture fluids as described previously (26,29). The FeLV (Gardner-Arnstein strain) used in this study represents a mixture of subgroups A and B.…”
