“…Nevertheless, our study showed that the promoter region of CYP1A1 contains three functional regions one proximal (-62 to -47) and two distal (-987 to -976 and -952 to -935) regions consistent with the results of a previous study reporting that an XRE sequence existed upstream of human CYP1A1 [53]. Both TCDD and omeprazole induced CYP1A1 transcription via the dephosphorylation of Sp1 at Ser-59 by PP2A [27]. In addition, the binding of Sp1 to the proximal promoter region of CYP3A4 has been shown [22], and Sp1 regulated both the basal and rifampin (RIF)-mediated transcription of porcine CYP3A46 by binding to its related cis-regulatory element in the proximal promoter and distal enhancer [56].…”

“…Sp1 and AhR/Arnt heterodimers bind to the Sp1‐like motif and XRE, respectively, and enhance the constitutive and inducible transcriptional activities of human CYP1B1 . Gut‐enriched Krüppel‐like factor inhibits Sp1‐mediated activation of the CYP1A1 promoter in Chinese hamster ovary cells , and TCDD induces CYP1A1 transcription via protein phosphatase 2A (PP2A)‐mediated dephosphorylation of Sp1 at Ser‐59 . However, the relationship between Sp1 and AhR in mediating the basal expression of CYP1A1 has not been systematically clarified.…”

“…The amino acid sequence for Sp1 in pigs and humans has more than 90% identity similarity, leading us to believe that Sp1 might have a conserved regulatory function in CYP1A1 expression. In humans, Sp1 activates the CYP1A1 promoter ( Zhang et al, 1998 ), and phosphorylation of Sp1 regulates CYP1A1 expression ( Shimoyama et al, 2014 ). We also found that the GC box at the proximal promoter of p CYP1A1 is the main cis -element that plays a regulatory role by recruiting Sp1.…”

“…Whole-cell extracts were prepared as described previously [21]. BV-2 cells were seeded at a density of 1.0×10 5 cells/35-mm dish (for whole-cell extracts) or at a density of 1.0×10 6 cells/100-mm dish (for cell fractionation).…”
