“…To demonstrate the contribution of macrophages to hernia regression, they were systemically depleted via intravenous administration of commercially available liposomes loaded with clodronate (dichloromethylene-bisphosphonate, Liposoma), a drug that selectively induces apoptosis of macrophages upon phagocytosis 15 , 16 . Macrophage depletion is expected to occur at around 24 h after intravenous administration, and circulating monocyte depletion is reported to peak at 18 h after administration 17 .…”

“…Macrophage depletion is expected to occur at around 24 h after intravenous administration, and circulating monocyte depletion is reported to peak at 18 h after administration 17 . However, repopulating monocytes can be recruited from the bone marrow within a relatively short time, so repeated administrations were performed for a more prolonged macrophage and monocyte depletion 16 , 18 . Two transiently depleted groups were analyzed to understand the macrophage contribution along the progression of hernia formation and regression.…”

“…5 mg/ml clodronate-liposome were produced for macrophage depletion. The in vivo macrophage depletion was performed as previous description with minor modification (Weisser et al, 2012). Briefly, clodronate or PBS liposomes (200 ll) were i.p.…”

“…To investigate the effect of choroidal-resident macrophages on myopia in a mouse model, 3-weeks-old C57BL/6 J mice were randomly divided into 2 groups and were injected intraperitoneally with clodronate or phosphate-buffered saline (PBS) liposomes. In the clodronate liposome group, after being engulfed by macrophages, the clodronate liposomes were degraded by lysosomal phospholipases, which released clodronate into the macrophages and induced apoptosis, thereby promoting macrophage depletion (40). The injection volume of clodronate liposome was 0.1 ml per 10 g of mice and the injection frequency was once every two days.…”
