Objective: Telomere-related genes (TRGs) play a critical role in various types of tumors. However, there is a lack of comprehensive exploration of their relevance in lung cancer. This research aimed to verify the relationship between TRGs gene expression and the prognosis of patients with lung adenocarcinoma (LUAD), as well as the prediction of drug treatment efficiency. Methods: A total of 2093 TRGs were acquired from TelNet. The clinical information including age, tumor stage, follow up and outcome (death/survival) and TRGs expression profile of LUAD were obtained from the patients in The Cancer Genome Atlas (TCGA) database and the Clinical Proteomic Tumor Analysis Consortium (CPTAC) database. The two databases were used to construct and verify a prognostic model based on the expression of hubTRGs. The tumor mutation burden, immune infiltration and subtypes, as well as IC50 prediction of multiple targeted drugs were also evaluated in TRGs-divided risk groups. Results: A total of 335 TRGs were significantly differentially expressed in LUAD as compared with normal control. Among them, 9 TRGs (ABCC2, ABCC8, ALDH2, FOXP3, GNMT, JSRP1, MACF1, PLCD3, SULT4A1) were finally identified as hubGenes and used to construct a TRG risk score. The TRG risk score showed favorable performance in constructing a prognostic nomogram in predicting survival of LUAD, and the ROC curves at 1, 3 and 5 years were plotted and the AUROC values were 0.743, 0.754 and 0.735, respectively. Higher TRGs risk score correlated with worse immune subtypes and higher tumor mutation burden in LUAD tissues. In addition, the patients in TRG high risk group harbored a lower TIDE score which indicated potentially better response to immunotherapy. Conclusion: This study proposed a broad molecular signature of telomere-related genes that can be used in further functional and therapeutic investigations, and also represents an integrated modality for characterizing critical molecules when exploring novel targets for lung cancer immunotherapy.

Background: Lung cancer is a global problem and the second most common cancer case for newly diagnosed cancers in 2020. Lung cancer is related to the inflammatory process, and symptoms of depression are often found in lung cancer patients. Improving the quality of life is one of the goals in advanced lung cancer treatment. This study aims to find the relationship between increased inflammatory markers such as neutrophil lymphocyte ratio (NLR), c-reactive protein (CRP) and depression with quality of life in patients with advanced non-small cell lung carcinoma. Methods: This was an observational analytic study conducted using a cross sectional design at Prof. Ngoerah Denpasar General Hospital (November 2022-August 2023). Univariate analysis presents data in the form of frequency, mean and standard deviation. Bivariate analysis was performed using the Chi-Square test and multivariate analysis using logistic regression. Results: The total samples in this research were 110 samples. The results of bivariate analysis showed that there was a significant relationship between NLR and CRP with depression (OR=3.225, p=0.005; OR=4.645, p=0.001), NLR and CRP with quality of life (OR=4.46, p=0.003; OR=5.313, p<0.001) and depression with quality of life (OR=31.385, p=<0.001). The results of multivariate analysis showed that NLR and CRP are independent risk factors for depression and decreased quality of life in patients with advanced non-small cell lung carcinoma. There was also a significant influence of age on the occurrence of depression (OR=0.364, p=0.037) and the influence of histology type (OR=0.493 p=0.039) on the quality of life of lung cancer patients. Conclusion: There was significant relationship between NLR and CRP with depression and quality of life in patients with advanced non-small cell lung carcinoma. Beside inflammatory markers, age is also related to the occurrence of depression and the type of histology influences quality of life.

Background Evidence suggests a mind-body component to aging, through which psychological distress from anxiety and depression drives molecular changes that promote early decline (ie, accelerated aging). Cancer survivors experience particularly high rates of anxiety and depression. Some survivors also have accelerated aging, though the relationships between anxiety/depression and aging are not clear. A synthesis of evidence is needed to understand the state of the science and impending priorities. Methods PubMed, Embase, CINAHL, Web of Science, and PsycNet databases were searched for studies that measured associations between depression, anxiety, and non-chronological aging in cancer survivors (2012 to 2022). Data were methodologically evaluated. Results Survivorship studies were included if they were peer-reviewed, published in English from 2012-2022, and measured associations between anxiety/depression and aging. 51 studies were included. Just over half were cross-sectional (53%). Foci included functional (n = 35, 69%) and biological (n = 16, 31%). Functional aging measures included frailty, sarcopenia, geriatric assessment, and cognition. Biological aging measures included telomere length, telomerase, age-related inflammatory blood-based biomarkers, renal insufficiency, anemia, and DNA methylation. 223 associations were tested. Associations between anxiety, depression and aging were generally positive, though with varying strengths. Most compelling were associations between functional aging-depression. There were concerns for selection and measurement biases. Conclusions Findings suggest positive associations between anxiety, depression and aging among cancer survivors. Future work is needed to clarify temporality, develop a consensus on the operationalization of aging, and diversify cohorts.
