BackgroundReference procedures are required for evaluating the accuracy of routine analytical systems for uric acid (UA). External quality assessment (EQA) for UA has only been conducted with quality controls in China, and the results have not been published. This study was designed to investigate both the trueness and inter-laboratory precision of UA measurements among routine analytical systems using a candidate reference method.MethodsWe performed the HPLC method recommended by the Japan Society of Clinical Chemistry (JSCC). Next, we evaluated its analytical performance and validated its trueness. The performance of 4 routine analytical systems (5 instruments per system, n=20) for UA was assessed by using 4 frozen pooled serum samples measured by the HPLC method according to biologically relevant quality goals.ResultsWithin-run, between-run, inter-day, and total CV of the method were less than 0.3%, 0.4%, 1.8%, and 2.6%, respectively. The UA measurements were consistent with the target values of standard reference material (SRM) 909b, the sixth ring trial for Reference Laboratories (RELA-2008) specimen, and national primary reference materials. The 4 frozen pooled serum samples were homogeneous, stable, and commutable. All routine systems achieved the desirable performance goal (total error <11.9%).ConclusionsWe successfully reproduced the JSCC's HPLC method, which was simple, specific, precise, and accurate. We recommend this method as a reference method for UA measurement in human serum. Four routine analytical systems for UA measurement had acceptable traceability, and their UA results showed good concordance.

This study demonstrates a highly stable, selective and sensitive uric acid (UA) biosensor based on high aspect ratio zinc oxide nanorods (ZNRs) vertical grown on electrode surface via a simple one-step low temperature solution route. Uricase enzyme was immobilized on the ZNRs followed by Nafion covering to fabricate UA sensing electrodes (Nafion/Uricase-ZNRs/Ag). The fabricated electrodes showed enhanced performance with attractive analytical response, such as a high sensitivity of 239.67 μA cm−2 mM−1 in wide-linear range (0.01–4.56 mM), rapid response time (~3 s), low detection limit (5 nM), and low value of apparent Michaelis-Menten constant (Kmapp, 0.025 mM). In addition, selectivity, reproducibility and long-term storage stability of biosensor was also demonstrated. These results can be attributed to the high aspect ratio of vertically grown ZNRs which provides high surface area leading to enhanced enzyme immobilization, high electrocatalytic activity, and direct electron transfer during electrochemical detection of UA. We expect that this biosensor platform will be advantageous to fabricate ultrasensitive, robust, low-cost sensing device for numerous analyte detection.

BACKGROUND: A short processing time and efficient scale-up stem cell isolation bioprocess is essential to exploit the potential of these cells for the treatment of multiple chronic diseases. Various methodologies have been used for stem cell recovery. However, most of them present economic and/or time-consuming drawbacks. RESULTS: This paper reports the characterization and optimization of immunoaffinity aqueous two-phase systems (ATPS), a liquid-liquid based separation technology enhanced with the PEGylation of the antibody as a promising platform in route tothe separation of CD133 + stem cells. The best results were obtained with Ucon-DEX or PEG-DEX systems with tie-line length (TLL) 15 or 20% w/w and volume ratio (V R ) 3. When the PEGylated antibodies were added to these selected optimized ATPS, they partitioned to both phases, but with higher preference for the bottom phase. CONCLUSION: PEGylation was an effective strategy to induce a change in the partition behavior of the CD133/2-biotin antibody when Ucon-DEX ATPS were used. Similarly, variation of the parameters (TLL and V R ) resulted to be useful for this goal.Nevertheless, further studies are required to find the optimal ATPS composition that will fractionate 100% of the antibody to the contaminants' opposite phase, making this system an ideal candidate to be tested for the selectivity of CD133 + stem cells.
