Betulin is the most popular of the known triterpenoids of the lupan series. It has valuable pharmacological properties. It exhibits antibacterial, antiviral, antitumor, hypolipidemic and other types of activity. The prospects of using betulin in medicine, pharmacology, and veterinary medicine require the development of effective methods for obtaining it from waste from the woodworking industry. Therefore, the question arises of the need to develop a technology for isolating and purifying triterpenoids from birch bark in large quantities. This review contains a variety of methods for the isolation of betulin. The advantages and disadvantages of the proposed methods are described. The following methods are considered: vacuum sublimation after preliminary alkaline; vacuum pyrolysis; supercritical extraction with carbon dioxide or mixtures of carbon dioxide with various solvents; extraction with organic solvents. Also, the method of microwave is described, it is activation on the example of the Kyrgyz birch (Betula kirghisorum), this is an endemic species that has not been studied before, growing on the territory of the Republic of Kazakhstan.

Natural origin products are regarded as promising for the development of new therapeutic therapies with improved effectiveness, biocompatibility, reduced side effects, and low cost of production. Betulin (BE) is very promising due to its wide range of pharmacological activities, including its anticancer, antioxidant, and antimicrobial properties. However, despite advancements in the use of triterpenes for clinical purposes, there are still some obstacles that hinder their full potential, such as their hydrophobicity, low solubility, and poor bioavailability. To address these concerns, new BE derivatives have been synthesized. Moreover, drug delivery systems have emerged as a promising solution to overcome the barriers faced in the clinical application of natural products. The aim of this manuscript is to summarize the recent achievements in the field of delivery systems of BE and its derivatives. This review also presents the BE derivatives mostly considered for medical applications. The electronic databases of scientific publications were searched for the most interesting achievements in the last ten years. Thus far, it is mostly nanoparticles (NPs) that have been considered for the delivery of betulin and its derivatives, including organic NPs (e.g., micelles, conjugates, liposomes, cyclodextrins, protein NPs), inorganic NPs (carbon nanotubes, gold NPs, silver), and complex/hybrid and miscellaneous nanoparticulate systems. However, there are also examples of microparticles, gel-based systems, suspensions, emulsions, and scaffolds, which seem promising for the delivery of BE and its derivatives.

Betulin-containing micronutrients increase the quality, physiological activity, and shelf life of food systems. Such micronutrients are relevant for developing physiologically functional food products for the population residing in the regions of the Arctic and the Far North. In this research, we aim to optimize the process of extracting betulin from Betula pendula Roth. For use in food systems for nutritional support of the population residing in regions with extreme climatic conditions by the method of mathematical modeling. Birch bark was harvested in the summer months of 2015–2021 in the foothill zone of the Altai Krai. Mathematical optimization of the process of betulin extraction from birch bark of white-trunked birches (Betula pendula Roth.), containing more than 20 wt% of the main substance, under autoclaving conditions was carried out using the Statistica 10 software package. Based on an analysis of response surfaces constructed using a regression equation, optimal parameters for betulin extraction with ethyl alcohol were determined. Thus, the birch bark particle size equals 3×3 mm; hydromodulus is 1:5; process duration is 150 min. Under these parameters, the betulin yield achieved more than 97%, with its authenticity and safety being confirmed experimentally. The physiological activity of betulin, as well as its usage examples in various food systems to prolong shelf life, prove its feasibility as a natural preserving agent. In addition, betulin can be used as a functional component, i.e., as an antioxidant for enriching food products, in particular, those intended for the population residing in extreme climatic conditions of the Arctic and the Far North.
