Deriving dopaminergic neurons for clinical use. A practical approach

Abstract: New small molecules that regulate the step-wise differentiation of human pluripotent stem cells into dopaminergic neurons have been identified. The steroid, guggulsterone, was found to be the most effective inducer of neural stem cells into dopaminergic neurons. These neurons are extensively characterized and shown to be functional. We believe this new approach offers a practical route to creating neurons of sufficient quality to be used to treat Parkinson's disease patients.

“…For example, 55 randomly selected homozygous pESC lines could completely match 80% of Japanese patients at three important HLA loci (A, B, and DR) [21]. Moreover, human pESCs could provide β cells, fibroblasts, cardiomyocytes, and neurons in vitro [20][21][22], suggesting that it might be possible to use these cells as an optimal source of genetically matched hESCs for cell therapy in female patients.…”

“…To generate authentic midbrain DA (mDA) neurons, we adopted dual SMAD inhibition and FP induction protocols . The common features of these protocols are early activation of Sonic Hedgehog (Shh and Pur) and WNT (ChIR) signaling during neural induction from hESCs by dual inhibition of SMAD signaling (SB431542 and LDN) (Fig.…”

“…Medium E has been used previously for embryonic stem cell culture, 15 and medium N has been used for neural stem cell culture. 16 Thus, media E and N may be able to provide a more similar environment to that found in vivo because HCECs in vivo are not in contact with serum, but instead face the aqueous humor produced by the ciliary body.…”
