Design and application of two oligonucleotide probes for the identification of Geodermatophilaceae strains using fluorescence in situ hybridization (FISH)

Abstract: Bacteria of the family of Geodermatophilaceae are actively involved in the decay processes [Urzì, C. and Realini, M. (1998) Int Biodeterior Biodegrad 42: 45-54; Urzì, C., Salamone, P., Schumann, P., and Stackebrandt, E. (2000) Int J Syst Evol Microbiol 50: 529-536] of stone monuments. Characterization of isolates includes phenotypic, chemotaxonomic and genetic analysis often requiring long-term procedures. The use of specific probes for members of Geodermatophilaceae family could be useful for the easy detecti… Show more

“…The focus, however, has been only on the identification of microorganisms living on stone monuments or wall paintings (Gurtner et al 2000;Urzí et al 2004;González and Saiz-Jiménez 2005). Moreover, little research has been conducted in the area of the biodegradative abilities of contaminating microflora, especially in relation to indoor artworks.…”

“…Representatives of the genus Geodermatophilus are characterized by the presence of a rudimentary mycelium, MK-9(H 4 ) as the major menaquinone and meso-diaminopimelic acid as the diagnostic diamino acid (Normand & Benson, 2012). Technical difficulties in culturing novel isolates (Urzì et al, 2004) hampered the study of the genus Geodermatophilus for many years, so that the type species, Geodermatophilus obscurus, and four subspecies whose names have not been validly published, 'G. obscurus subsp.…”

“…However, in spite of the already-demonstrated analytical potential [11,14] and versatility of RNA-FISH technique for unveiling potential biodeteriogens of CH [7][8][9][10][15][16][17][18][19][20][21][22], it continues to be scarcely applied in this field.…”

“…The RNA-FISH protocols already applied for investigating biodeterioration of CH has been performed in situ (directly in the material) [ 10,16,17] or ex situ (in coated-slides or tape-strip) [7][8][9][16][17][18][19][20][21][22] for investigating biodeterioration of a wide variety of materials (stone, rock, glass, mats, metals, mortars, polymers, wood, among others) [7][8][9][10][15][16][17][18][19][20][21][22], employing almost exclusively paraformaldehyde as fixative and analyzing the cells by epifluorescence microscopy or Confocal Laser Scanning Microscopy (CLSM).…”
