Design and Characterization of Spray-Dried Chitosan-Naltrexone Microspheres for Microneedle-Assisted Transdermal Delivery

Abstract: Naltrexone (NTX) hydrochloride is a potent opioid antagonist with significant first-pass metabolism and notable untoward effects when administered orally or intramuscularly. Microneedle (MN)-assisted transdermal delivery is an attractive alternative that can improve therapeutic delivery to deeper skin layers. In this study, chitosan-NTX microspheres were developed via spray-drying, and their potential for transdermal NTX delivery in association with MN skin treatment was assessed. A quality-by-design approach … Show more

“…Higher feed viscosity leads to the formation of larger droplets, and as a consequence, particles obtained during the spray drying process were characterized by larger diameters. A comparable result was observed in work of Ogunjim et al [ 31 ], where the spray drying of different chitosan solutions with low (0.5% w /v ), medium (0.7% w /v ) and high (0.9% w /v ) concentrations significantly affected microspheres size.…”

“…Spray drying is a continuous and scalable drying process to encapsulate materials, the process can generate nano to micron size particles that have a narrow distribution in a very short time. [108] It has been used in pharmaceutical processing [109][110][111] and food industry [112] for a long time. The main advantage of spray drying is the waste reduction and high efficiency of raw material.…”

“…Dried samples were mounted on double-sided adhesive carbon tabs mounted on Scanning Electron Microscopy (SEM) stubs and coated with gold/palladium and viewed under a JEOL 5600 (JEOL, Tokyo, Japan) scanning electron microscope at an accelerating voltage of 20 kV. Microcapsule size was determined using ImageJ (1.52 a; ), (accessed on 15 March 2022) resulting from the average of 400 measurements [ 38 , 39 ].…”
