Design and Synthesis of Spirocycles

Abstract: In this account we disclose a variety of simple strategies demonstrated in our laboratory in the last two decades for the assembly of diverse and intricate molecular frameworks containing spiro linkage(s) by using various metathesis protocols such as ring‐closing metathesis (RCM), ring‐opening cross‐metathesis (ROCM), ring‐closing enyne metathesis (RCEM) and ring‐rearrangement metathesis (RRM). Also, cycloaddition reactions ([2+2], [4+2] and [2+2+2]) and other key processes such as Claisen rearrangement (CR), … Show more

“…A pericyclic [4+2]‐cycloaddition protocol is a powerful tool and has been continuously used by the synthetic organic chemists for the generation of diverse medicinally active molecules. Since its finding in 1928 (Nobel Prize 1950) in the laboratory of Otto Diels, its applications in organic synthesis is growing exponentially day‐by‐day, and to my best knowledge almost every architecturally complex natural or non‐natural products involve this powerful tool at some stage to showcase the target molecules . In this context, a variety of dienes and dienophiles either commercially available or easily prepared have been employed in DA‐reaction .…”

“…They not only play momentus role in optoelectronics, displays and silver free imaging systems but also in bioactive naturals and non‐natural compounds. Keeping this in mind, spirotruxene derivatives 54 a‐f along with their photophysical properties have been reported by Kotha and his coworkers using cyclotrimerization, ring‐closing metathesis and Suzuki‐Miyaura cross‐coupling reactions as key transformations (scheme ) …”

“…In 2009, we have demonstrated the synthesis of highly substituted diphenylalkane derivatives via CEM under ethylene followed by the DA reaction and subsequent aromatization sequence. [74] The strategy involves a step-wise functionalization of acetylene moiety, where initial CEM with ethylene in the presence of the G-II catalyst generated the required diene. Then, the resulting diene was subjected to DA reaction followed by aromatization to yield highly functionalized aromatic frameworks.…”
