Design and validation of a diffuse reflectance and spectroscopic microendoscope with poly(dimethylsiloxane)-based phantoms

Abstract: Many cases of epithelial cancer originate in basal layers of tissue and are initially undetected by conventional microendoscopy techniques. We present a bench-top, fiber-bundle microendoscope capable of providing high resolution images of surface cell morphology. Additionally, the microendoscope has the capability to interrogate deeper into material by using diffuse reflectance and broadband diffuse reflectance spectroscopy. The purpose of this multimodal technique was to overcome the limitation of microendosc… Show more

“…Ongoing development work with microendoscopic systems may enable spectroscopic data to be used in conjunction with quantitative image features, such as those described in this paper. 45 This quantitative data, coupled with extracted imagebased features provided by algorithms such as QFEA, could lead to the adoption of standardized, objective methods of in vivo high-resolution microendoscopy image interpretation and analysis.…”

“…The first objective of this study was to design the multimodal instrumentation and associated contact fiber-bundle probe to co-register qualitative image data with quantitative spectroscopy data 29 53 . For the high-resolution fluorescence imaging modality, a 455 nm LED (20 FWHM) light source (Philips, USA) is coupled through a 1 mm-diameter image fiber (FIGH-50-1100N, Myriad Fiber Imaging, USA) consisting of approximately 50,000 individual 4.5 μm-diameter fibers.…”

The distal blue 455 nm LED light excites a contrast agent, such as proflavine, which emits fluorescence signal (peak emission of ~515 nm with quantum efficiency of ~0.5) back into the image fiber and is delivered to an 8-bit monochrome CMOS camera (FL3-U3-32S2M-CS, Point Grey, Canada) 29 53 60 . A filter set (Chroma Technology Corp., USA) separates the excitation and emission signals.…”
