Design of field trials for the evaluation of antibacterial products for therapy of bovine clinical mastitis

Abstract: In this paper, the design and statistical analysis of field trials for the evaluation of the efficacy of clinical mastitis therapeutics is covered. First, general issues underlying the design of clinical trials are reviewed. These include bias and confounding; randomization and blocking; and study objectives and choice of the corresponding hypothesis. Specific issues in the design of clinical mastitis trials are also discussed. Selection of subjects is discussed with regard to choice of experimental units, ide… Show more

“…Well‐designed prospective and controlled field trials are necessary in order to assess the efficacy of defined antimicrobial treatments for clinical mastitis (Schukken & DeLuyker, ; Barlow, ; Roberson, ). Most published studies lack an untreated control group, and many do not diagnose causal agents at species level.…”

“…Special care should be taken to prevent bias and confounding, and to ensure proper randomisation, blocking and statistical analysis (Schukken and Deluyker 1995). The effect of NSAID on recovery has been reported by others (1994, 1996).…”

“…The absence of a suitable pharmacokinetic model for the intra‐mammary antibiotics precludes optimization of therapy by pharmacokinetic–pharmacodynamic techniques (Nielsen & Friberg, ). Therefore, development of effective dose regimens for intra‐mammary drugs is dependent on large and expensive clinical trials (Schukken & Deluyker, ). Furthermore, veterinary clinicians and scientists are constrained in their decision‐making regarding mastitis treatments by the requirement to provide reliable predictions of drug residues in milk, as label instructions only apply to label conditions (Bourin & Clénet, ).…”
