Design of in situ porcine closed-circuit system for assessing blood-contacting biomaterials

Abstract: The overall pre-clinical process of determining the blood compatibility of any medical device involves several stages. Although the primary purpose is to protect the patients, laboratory testing has been over-utilized for many years with a huge number of unnecessary animal tests being done routinely. Recently, the elimination of needless testing has become important in controlling the cost of healthcare and in addressing many issues related to the ethics of animal research. With this in mind, we designed a new… Show more

“…Our data point to the positive role that peptide 2 can play in promoting EC coverage of the ePTFE surface. In addition, our previous in vivo study confirmed the improved hemocompatibility of the ePTFE‐2 surface. Future work should focus on studying the high EPC selectivity of peptide 2 more deeply.…”

“…The linear heptapeptide (Ph.D7) phage display libraries kits (New England Biolabs, Beverly, MA) were used for the in vitro biopanning experiments. Biopanning was conducted following a modified protocol from New England Biolabs (Ipswich, MA), as previously described . The following abbreviations for the peptides sequences were used: (1) GQSEKHL; (2) HGGVRLY; (3) SLSKWSF; (4) KIAVIST; and (5) VSRDTPQ.…”

“…155 In another study, layered PCL and PGA composite sheets seeded with bovine fibroblasts, SMCs, and ECs for up to 2 weeks showed a similar stress-strain response to that of bovine arteries. 135 The unpredictable transition of mechanical behavior of synthetic TEVGs over time as the scaffold degrades requires a long-term in vivo study with appropriate assays at each time point to observe the graft mechanics regarding degradation. “…Munisso et al [14] developed of a simple in situ porcine closed-circuit system for assessing blood contacting biomaterials to explore the interaction between several materials and blood. This system allowed large numbers of materials (up to 16) to be simultaneously evaluated using the fewest number of animals.…”
