Design of Peptide Ligand for Lactoferrin and Study of Its Binding Specificity

Abstract: The in silico modelling of peptides complementary to lactoferrin was carried out using the Protein 3D software package and replication of the natural bonding site between pneumococcal surface protein (PSP) and lactoferrin (LF). The modeling was based on analysis of the conjugated ion–hydrogen bond systems between these proteins (CIHBS). The oligopeptide EEVAPQAQAKIAELENQVHRLE was proposed via computer modelling and synthesized using the solid phase synthesis technique, purified, and analyzed with MS and HPLC m… Show more

“…The electrophoretic homogeneity of synthetic peptide containing 21 AA residues proposed as lactoferrin binding ligand by in silico modeling was confirmed by CE analysis in BFS capillary (75 µm id, 60/50 cm total/effective length) thermostated at 30°C using 10 mM Tris/HCl buffer, pH 7.2, as BGE, 20 kV as separation voltage, and 200 nm as detection wavelength [229]. The CE analysis of a mixture of this peptide with lactoferrin proved formation of peptide–lactoferrin complex.…”

“…Peptides have a number of advantages over more widely used antibodies and aptamers, such as low cost, ease of synthesis and modification, high stability and regenerability, low immunogenicity and toxicity, and the ability to be designed as needed [228]. However, peptides also have a number of disadvantages, such as low affinity and specificity compared to antibodies, low catalytic activity compared to enzymes, and the potential for denaturation, aggregation, and degradation under certain conditions.…”
