Design of Poly(lactic-co-glycolic Acid) (PLGA) Nanoparticles for Vaginal Co-Delivery of Griffithsin and Dapivirine and Their Synergistic Effect for HIV Prophylaxis

Abstract: Long-acting topical products for pre-exposure prophylaxis (PrEP) that combine antiretrovirals (ARVs) inhibiting initial stages of infection are highly promising for prevention of HIV sexual transmission. We fabricated core-shell poly(lactide-co-glycolide) (PLGA) nanoparticles, loaded with two potent ARVs, griffithsin (GRFT) and dapivirine (DPV), having different physicochemical properties and specifically targeting the fusion and reverse transcription steps of HIV replication, as a potential long-acting microb… Show more

“…Therefore, results of in this study indicate that release of CVL from NPs is not predominantly driven by a solo mechanism, but a combined mechanism of Fickian and non-Fickian release. When the literature is examined, similar results are found [42].…”

“…PLGA NPs can be formulated by use of emulsion evaporation, emulsion diffusion, solvent displacement and nanoprecipitation techniques. Figure 2 shows the use of a double emulsion-solvent evaporation technique in the formulation of a PLGA based co-delivery platform [31]. Poly(lactic acid-co-glycolic acid) (PLGA) is a polyester, copolymer block made up of polyglycolic acid and poly-lactic acid [28].…”

“…This highlights the need to develop prevention systems that women can initiate without their partner's consent [2]. In this scenario, topical preexposure prophylaxis (PrEP) with antiretroviral drugs is one option to prevent the sexual transmission of HIV, since they can be applied in the vagina and serve as a method of protection that can be controlled by women themselves, thus empowering them in the fight against HIV-1 infection without the consent of their sexual partner [3].…”
