“…coli AG100 tet ( acrB ++ ) (Figure A) and P. aeruginosa BAA-2795 (Figure B) was negligible as compared to that of untreated cells. In contrast, PAβN and polymyxin B (Gram-negative outer membrane permeabilizer) treated cells showed enhanced fluorescence. Thus, the synergism of the RP1 with the antibiotics observed was not due to the off-target effect of outer membrane permeabilization.…”

“…This result is not surprising given the potentiation effects thioridazine (35) showed with the knockout as well as the wild-type and its antimicrobial activity against the knockout strain. Despite trifluoperazine (37) sharing these characteristics, it did not appear to damage the outer bacterial membrane as all other compounds besides thioridazine (35) showed similar results to the nitrocefin-only control (Figure 3).…”

“…Antifungal drugs both butenafine (19) and naftifine (27) showed a 2-fold reduction in MIC with the knockout strain at 16 and 32 μg/mL, respectively, suggesting an additional mechanism of antimicrobial potentiation. Finally, the antipsychotics both thioridazine (35) and trifluoperazine (37) showed a 4-fold reduction in erythromycin MIC at 8 μg/mL as well as showed an antimicrobial activity alone against the knockout strain with the same MIC (32 μg/mL). This suggests another mechanism in addition to efflux inhibition and that these compounds are also substrates of AcrB as they did not have antimicrobial activity alone in the pump-active wild-type strain.…”
