Silk sericin (SS), a protein biopolymer, is gaining an increasing attention in a variety of biomedical applications. Our objective was to identify the optimal extraction method yielding the highest silk protein content, to assess its chemical elemental composition, to characterize it, and to compare its conformation by X‐ray diffraction and Fourier‐transform infrared versus that of the commercial one. Herein, alkali and high temperature is the perfect approach. This study aimed further to test whether extracted SS could attenuate the adverse hepatotoxic effects induced by diethylnitrosamine (DEN) in mice. Animals were injected intraperitoneally once weekly for four consecutive weeks with physiological saline (0.9% NaCl), DEN (25 mg/kg), SS (1 g/kg) 90 min prior to DEN injection, melatonin (10 mg/kg) followed within 1 h with the administration of SS and 30 min later with DEN. The results confirmed the hepatotoxicity of DEN manifested by an increase in liver enzymes' activity, and pathological changes in the liver tissue. The combined use of SS and/or melatonin with DEN returned most of the parameters to values similar to the controls. Briefly, chemically characterized SS seems a valuable extract for further consideration as anti‐carcinogenic agent. It has the potential to upgrade liver cancer chemotherapeutic agents.

We report the feasibility of electrospinning of protein-polymer multilayered scaffolds with selected materials such as non-hydrolyzed silk sericin (SS), polyethylene oxide (PEO), and polylactic acid (PLA), with tuned fiber size and properties for each layer. We present a new innovative way for the electrospinning (ES) of non-hydrolyzed SS mixed with PEO yielding fibers with an average diameter ranging between 120 and 150 nm. Different SS:PEO ratios have been electrospun to study the effect of the concentration of SS protein on the fibers size and shape, as well and their electrospinnability. Electrospun SS:PEO fibers display weak to no mechanical resistance (non-measurable) and their deposition onto a sturdier scaffold is necessary to allow their use in biomedical and/or pharmaceutical fields. Therefore, bilayer scaffolds have been fabricated consisting of a PLA support and SS:PEO fibers obtained from the optimized SS:PEO ratio (1.2:4). They are composed of a sturdy hydrophobic layer of PLA fibers and a layer of sticky hydrophilic SS:PEO fibers. The scaffolds have been characterized extensively by Fourier transforms infra-red (FTIR) spectroscopy, X-ray diffraction (XRD), scanning electron microscopy (SEM), and their resistance to mechanical stress. Finally, hydrophobicity of both layers has been determined by measuring the contact angle of water droplets on the scaffolds, further proving the bilayer nature of the scaffolds.

Biorefinery, the process of converting biomass to valueadded materials, is gaining significant interest because of the depletion of natural resources and an increasing awareness of the need for sustainable development and material production. Silk sericin, an abundantly available byproduct of the sericulture industry, has been identified for biorefinery usage since it is biodegradable, bioavailable, pH and temperatureresponsive, naturally photoluminescent, has antioxidant properties, and is a GRAS and FDA-safe food additive. This Perspective examines the properties of sericin and its sources and discusses currently explored applications, including as a delivery system for drugs or genetic material, for wound healing, tissue engineering, media supplements, cosmetology, packaging materials, edible coating, and as bio adhesives. This Perspective examines the sources of sericin, its properties, and some of the currently explored applications. These include biomedical applications such as a delivery system for drugs or genetic material, wound healing, tissue engineering, media supplements, cosmetology, packaging materials, edible coating, and as bio adhesives. Finally, we highlight the recent developments in sericin modification to functionalize it for specific applications; this is a promising future direction that will further enhance the potential use of this valuable biomass.
