Detection and characterization of urinary metabolites of boldione by LC‐MS/MS. Part I: Phase I metabolites excreted free, as glucuronide and sulfate conjugates, and released after alkaline treatment of the urine

Abstract: Boldione (1,4-androstadiene-3,17-dione) is included in the list of prohibited substances, issued by the World Antidoping Agency, in the group of exogenous anabolic steroids.Endogenous production of low concentrations of boldione has also been reported. The objective of the study was to assess boldione metabolism in humans. Detection of Metabolite M7 was identified as the 5β-isomer of 1-androstenedione, and metabolites M1 to M4 were hydroxylated metabolites and tentative structures were proposed based on mass … Show more

“…) and their analytical conditions are listed in Table 1. Metandienone and boldenone sulfate metabolites, MEDm-S [7] and eBOLD-S [5,12], were included. For stanozolol, three recently reported glucuronides STAN-N-G, STAN-O-G and eSTAN-N-G, together with 4STAN-G and 16STAN-G were incorporated to the method [14].…”

“…Boldenone, boldione, methyltestosterone and metandienone metabolites conjugated with sulfate have been recently identified [5][6][7][8]12]. Glucuronoconjugated metabolites poorly or not hydrolyzable using ␤-glucuronidases have been described for testosterone and stanozolol [13,14].…”

“…A peak at m/z 518 present in the samples post-administration and not present The presence of androst-1,4,6-triene-3,17-dione and its derivatisation artefact as described by Parr et al [24] after androst-1,4,6-triene-3,17-dione administration was also detected Table 2. Boldenone and boldenone metabolite measurements on urine samples collected after the administration of 100 mg of boldione. The reported concentrations were estimated from the GC-MS presuntive screening analysis after the formation of the corresponding TMS derivatives as boldione metabolite, while Gómez et al [8] described this as boldione metabolite only after alkaline hydrolysis of the sample. The derivatization artefact of the 17-reduced metabolite, androst-1,4,6-triene-17β-ol-3-one, was also identified.…”

“…The suitability of these methods for the open detection of AAS with unknown structure allowed for the discovery of several (minor) previously unreported metabolites [43][44][45][46][47][48]. Currently, the suitability of a precursor ion scan method [39] for the routine screening of unknown AAS is being tested in our laborato ries.…”
