Detection and Structural Characterization of SFAHFA Homologous Series in Mouse Colon Contents by LTQ-Orbitrap-MS and Their Implication in Influenza Virus Infection

Abstract: Fatty acid esters of hydroxy fatty acids (FAHFAs) are a new class of endogenous lipids with promising physiological functions in mammals. We previously introduced a new type of lipids to this family called short-chain fatty acid esters of hydroxy fatty acids (SFAHFAs), branching specific to the C2 carbon of a long-chain fatty acid (≥C20). In this study, we discovered a homologous series of SFAHFAs comprising C16–C26 hydroxy fatty acids esterified with short-chain fatty acids (C2–C5) in mouse colon contents. Th… Show more

“…Compared with traditional high‐performance liquid chromatography (HPLC), UHPLC improves the efficiency of chromatography and shortens the analysis time, while Q‐Exactive Orbitrap‐MS produces accurate mass measurements and rich snippets of information. Therefore, UHPLC‐Q‐Exactive Orbitrap‐MS, supports the valid component analysis of plant extracts 24 . Second, the potential targets and pathways of BYD in IPF treatment were predicted through network pharmacology.…”

“…Finally, their structures were characterized by nuclear magnetic resonance (NMR) spectroscopy and high-resolution electrospray ionization mass spectrometry (HR-ESI-MS). The linear ion trap quadrupole-Orbitrap mass spectrometry was used to acquire HR-ESI-MS spectra by the conditions mentioned in our earlier report [ 24 ].…”

“…Lipid separation was achieved using an Atlantis T3 C18 column (2.1 mm × 150 mm, 3 μM, Waters, Milford, MA), with a mobile-phase flow rate of 0.2 mL/min (A: aqueous 10 mM CH 3 COONH 4 , B: isopropanol, C: methanol) at 40 °C of column temperature throughout the analysis. These LC/MS conditions were consistent with our earlier research . The mobile phase composition was adjusted as follows: 30% B and 35% C from 0 to 1 min, 75% B and 15% C from 1 to 9 min, 82.5% B and15% C from 9 to 21 min, 95% B and 5% C from 21 to 25 min, 30% B and 35% C from 25 to 26 min, and 30% B and 35% C from 26 to 30 min.…”
