Detection of a novel HLA‐B27 allele, B*2740, in Taiwanese volunteer bone marrow donors by sequence‐based typing: curiosity rewarded

Abstract: We report here a novel HLA-B allele, B*2740, discovered in Taiwanese volunteer marrow donors. The new sequence has nucleotide variation at position 527 (T-->A) as compared to B*2708. The nucleotide change caused an amino acid substitution from valine (V) to glutamic acid (E) at codon 152. Since B*2740 carries sequence confers to HLA-Bw6 public epitope we believe that this novel B*27 allele might have been generated from a gene conversion involving a Bw4-specific allele (probably B*2704) and a Bw6-specific alle… Show more

“…Thus, the database of Taiwanese bone marrow donor registry consists of HLA alleles and haplotypes with its uniqueness. From time to time, new alleles, rare frequency alleles and Taiwanese‐conserved haplotypes were identified in our routine HLA typing practices (Chen et al. , 2010; Yang & Lin, 2007; Yang et al.…”

“…B*27:40 was accidentally discovered by an attempt to validate its certainty using SBT method on all donors who were thought to carry B*27:08, an HLA‐B allele found in Caucasians. From our experience, we suggest oriental individuals who were previously tested to express B*27:08 by RELI‐SSO method should be re‐evaluated by SBT for its accuracy (Chen et al. , 2009c).…”

“…The DNA material was subjected to HLA genotyping for HLA-A,-B,-C,-DRB1, and-DQB1 loci using a commercial polymerase chain reaction- sequence-based typing (PCR-SBT) kit, the SeCore ® A/B/C/DRB1/DQB1 Locus Sequencing kit (Life Technologies, Brown Deer, WI, USA). High-resolution allelic sequencing was performed as previously described [1011]. …”
